Large-scale Genotyping Service

The Affymetrix GeneChip® Human Mapping 10K Array Xba 142 2.0 provides comprehensive single nucleotide polymorphism (SNP) genotyping across the human genome at a median intermarker distance of 105 kb. Only 250 ng of genomic DNA are required to obtain more than 10.000 genotypes.

The Affymetrix GeneChip® Human Mapping 100K Set is comprised of two arrays, each with greater than 50.000 SNPs. One array uses the XbaI restriction
enzyme, while the second uses HindIII. With 100.000 SNPs obtained in a single experiment at a median intermarker distance of 8.5 kb, the Mapping 100K Set is  the first in a family of products for whole-genome association studies (Comparison of Mapping 10K and 100K SNP coverage to 400 marker microsatellite panel).

The GeneChip® Mapping products are the ideal tools for:

  • Linkage analysis
  • Identification of chromosomal copy number changes
  • Linkage disequilibrium studies
  • Cancer and population genetics
  • Whole-genome association studies (Mapping100K Set)

The Large-Scale Genotyping Service follows this general workflow (technology and graphical overview - example given for the Mapping 10K 2.0 Array):

  • Advice on suitable experimental strategy, DNA extraction etc.
  • Restriction digest with Xba
  • Adapter Ligation
  • One-primer PCR amplification
  • Fragmentation and End Labeling
  • Hybridization to GeneChip® Human Mapping 10K 2.0 Array
  • Staining and washing of array
  • Scanning with confocal laser scanner
  • Quality check of raw data using Affymetrix software (GCOS 1.1 and GDAS 3.0)
  • Computation of genotype calls (GDAS 3.0)
